assessing the genetic diversity of tobacco (nicotiana tabacum l.) varieties
Authors
abstract
the genetic diversity of 40 tobacco (nicotiana tabacum l.) cultivars was analyzed using 12 issr primers. thecultivars were evaluated for 14 traits using a simple lattice design (7×7) with two replications. of 149 amplified bands, 142 (95.3%) were polymorphic. maximum and minimum bands were generated by primers ubc825 (17 bands and 94.11% polymorphic) and ubc824 (7 bands and 100% polymorphic), respectively. of the studied primers, ubc824 showed maximum polymorphism information content (pic) (0.43) and the highest genetic diversity, and thus could be used in future genetic diversity studies. in principal coordinate analysis using a similarity matrix, the first two coordinates, which are the main coordinates, explained 41.97% of the total variance. cluster analysis was performed to determine the genetic relationship of tobacco cultivars using the un-weighted pair-group method with arithmetic average (upgma) based onmorphological traits and issr markers separately. plant genotypes were divided into nine main groups of flue-cured tobacco cultivars, and a cophenetic correlation coefficient was calculated (r = 0.9), indicating the usefulness of the upgma method for clustering plant genotypes. cluster analysis based on morphological traits divided the studied genotypes into five groups using the upgma method. results of canonical discriminate function analysis using the fisher linear method showed that the upgma method separated the genotypes with 100% accuracy. both clusters were consistent with the geographical origins of the cultivars.

Coproporphyrinogenase in tobacco (Nicotiana tabacum L.).
1. Coproporphyrinogenase was extracted and purified from tobacco (Nicotiana tabacum L.). Enzyme activity was mainly located in mitochondria rather than in chloroplasts. The enzyme was purified by differential centrifugation, ammonium sulphate fractionation, calcium phosphate gel adsorption and dialysis. A 69-fold final purification was obtained. 2. An apparent K(m) value of 3.6x10(-5)m was foun...
full textGenetic diversity among flue-cured tobacco (Nicotiana tabacum L.) revealed by amplified fragment length polymorphism
Flue-cured tobacco (Nicotiana tabacum L.) is one of the most important commercial crops in the world. Genetic diversity studies provide estimates on the level of genetic variation among diverse materials that can be used in germplasm management, varietal protection, and flue-cured tobacco improvement. Amplified fragment length polymorphism (AFLP) analysis of 51 flue-cured tobacco cultivars prod...
full textGenetic Study of Agronomic Traits in Tobacco (Nicotiana tabacum L.) Using Hayman’s Graphical Analysis of Diallel
For the purpose of graphical analysis and estimation of genetic parameters related to yield and its components, six tobacco genotypes was crossed in half -diallel mating design. In this study, the F2 progenies and their parents were evaluated in a randomized complete block design with three replications. The results of analysis of variance indicated a significant difference between genotypes fo...
full textBiotechnological Reduction of Tobacco (Nicotiana Tabacum L.) Toxicity
BACKGROUND: Nicotiana tobacco contains large amounts of alkaloid nicotine. Tobacco plant is used for smoking and causes many health problems since it is high in nicotine which is one of the widely-recognized toxic compounds with serious side effects for different body organs. Reducing nicotine content of this plant is a way to reduce its health hazards in cigarette smokers. Utilizing the new ...
full textMonograph of Tobacco (nicotiana Tabacum)
The use of tobacco dates back to the ancient civilizations of the Americas, where it played a central role in religious occasions. The peoples smoked tobacco in cigars and pipes and chewed it with lime, for its pleasurable euphoriant effects. In the 16 th century, Europeans spread the use of tobacco in North America, while the Spanish bring it into Europe. In the 1559, Jean Nicot, the French am...

Tobacco (Nicotiana tabacum L.) has become a model system for tissue culture and genetic engineering over the past several decades and continues to remain the 'Cinderella of Plant Biotechnology', An ill vitro culture medium (Murashige and Skoog, 1962), based on the studies with tobacco tissue cultures, has now been widely used as culture medium formulation for hundreds of plant species. Studies ...
